The p24 kit
The p24 kit is an assay designed to detect the p24 protein, a core protein of HIV-1, in various biological samples such as blood, serum, or plasma. The p24 protein is a component of the HIV-1 viral capsid and is commonly measured to monitor HIV-1 infection, particularly in early stages or during antiretroviral therapy (ART).
Key Components and Functionality of a p24 Kit
- Capture Antibodies: The kit includes wells coated with monoclonal antibodies specific to the HIV-1 p24 antigen. These antibodies are designed to bind the p24 protein present in the sample.
- Sample Addition: The biological sample suspected of containing the HIV-1 virus (e.g., serum, plasma) is added to the wells. If p24 protein is present, it binds to the capture antibodies on the well surface.
- Detection Antibodies: After the sample is incubated, a secondary antibody specific to a different epitope of the p24 protein is added. This antibody is conjugated to an enzyme, typically horseradish peroxidase (HRP), or a fluorescent label, depending on the detection method.
- Signal Development: For enzyme-linked assays (ELISA), a substrate is added that reacts with the enzyme to produce a measurable color change. For fluorescent-based assays, a fluorescent signal is emitted upon binding. The intensity of this signal is proportional to the amount of p24 protein in the sample.
- Quantification: The signal is measured using a spectrophotometer (for ELISA) or a fluorometer (for fluorescent assays). The concentration of p24 is determined by comparing the sample's signal to a standard curve generated from known concentrations of p24 protein.
